The RPA kit used in this workflow appears in a study published in Irish Veterinary Journal.
Bovine respiratory disease (BRD) remains a major challenge in cattle production. Identifying bacterial pathogens such as Pasteurella multocida often depends on laboratory-based methods, which can limit access to timely results in resource-limited settings.
A recent study by researchers at the College of Veterinary Medicine, Qingdao Agricultural University, demonstrates the potential of an RPA-CRISPR/Cas12a workflow for on-site pathogen detection. Targeting kmt1, a species-specific gene of P. multocida, the researchers developed an assay combining recombinase polymerase amplification (RPA), CRISPR/Cas12a detection, and lateral flow strip (LFS) readout.
RPA amplification was completed in 15 minutes at 37 °C. Following CRISPR/Cas12a detection, results could be visually interpreted using a lateral flow strip, without requiring a thermal cycler or fluorescence reader.

Study Highlights
The study reported a detection limit of 10 copies/µL, with no cross-reactivity against ten other tested bovine pathogens, including Mannheimia haemolytica, Histophilus somni, and bovine viral diarrhea virus.
In a blinded validation against conventional PCR, the assay showed complete agreement across 40 archived bovine nasal swab samples collected from cattle with clinical signs of BRD at commercial farms (17 positive, 23 negative).
In a separate validation involving 42 clinical nasal swab samples, the assay also showed 100% agreement with real-time qPCR, with reference Ct values ranging from 16.0 to 35.0 (36 positive, 6 negative).
These data describe the complete detection workflow developed in the study, not the performance of any single reagent component within it.
